“We know that everyone needs a personal space. But how large is a personal space required to be?” Is the second sentence grammatically correct? And is it semantically appropriate?

Are you talking about a place or a concept? “Personal space” usually refers to the “bubble” around a person that shows how physically close others can be to that person. “A personal space” sounds more like a place, like your room or a treehouse, where you enjoy being.

OK, let’s change it to “personal space.” What do you think about the second sentence?

Then I don’t think “large” really fits, as a concept usually can’t be large. Ummm…maybe “How much personal space is really required?” or something like that.
