In terms of applying to schools like USC, JHU, WUSTL, Duke and others, does a Math II subject test really help or will a high score on, lets say, Bio and US History do fine?

They’re all the same. It’s not like Math II is more important than Bio.

USC: recommends but does not require IIs, taking Math 2 is a good idea for applying for science or engineering but otherwise any two will do.

JHU: does not require but recommends three IIs (so you should seriously consider taking a third one for there); as to engineering it virtually requires at least two IIs because it says you “should” submit a Math 2 and a science; otherwise math not needed

Duke requires two IIs: math 2 is required for engineering, otherwise not needed.

WUSTL: recommends two IIs without specification but if applying engineering or science you should consider it.

drsuba is right in suggesting it depends on what you want to study. For engineering or science it is better. For history, it does not matter.
