I play a traditional Tibetan instrument called the dranyen, and I am wondering if it would be appropriate to include a video of me playing the dranyen as part of my music supplement. College Admissions Officers usually expect western instruments, but I think this would definitely make me stand out. Another question is I also play two other instruments native to Tibet. How should I submit multiple videos of me playing different instruments?

You have a point about it making you stand out; it would be better if it made you stand out in a way that’s relevant to what you want to do though. So, what do you want to do?
As to your second question, can you not do a short video with segments of you playing all three?
@Philpsych I want to focus on the biological sciences, but I have a strong background in music. I have been playing these three instruments at a young age, I think I began around 7. In addition, I’m also part of my high school band, I play the trumpet. Music is more like my hobby/interest.
I see. Well, if it’s not something you’re deeply passionate about to the point where it’s an essential part of you, I would advise you not to send a supplement.
I disagree with the above commenter- it is truly unique, and it could add to your application. If the college allows supplemental material, I would strongly recommend you sending in a supplement.
