The problem is when I filled up my I-20, I accidentally wrote a wrong date and then tried to rewrite it. Now, my I-20 has a pretty visible blot on “Date of sign” section. Should I be worried about it and can it be by any chance the reason for council to deny my visa?

Couldn’t you have used white-out or something instead of trying to rewrite it in pen?
Definitely don’t use whiteout on immigration paperwork.
My college always had me fill out a new form whenever I made a mistake on government paperwork. I don’t know how problematic it would actually be in practice.
If you have time before your visa interview, you can request a new I-20 from your college. If you don’t have time, just use the form you have. Worst case they’ll make you come back with a clean form.
